PURPOSE: Provided are a method and a kit for simultaneously detecting both Plasmodium falciparum and Plasmodium vivax by one-shot multiplex PCR, thereby rapidly and correctly detecting both Plasmodium falciparum and Plasmodium vivax. CONSTITUTION: The method for simultaneously detecting both Plasmodium falciparum and Plasmodium vivax comprises performing multiplex polymerase chain reaction(PCR) of a gene encoding 18s rRNA of both Plasmodium falciparum and Plasmodium vivax, using a genus specific sense primer and a species specific antisense primer, wherein the sense primer has a nucleotide sequence selected from SEQ ID NOs 1 to 17; and the antisense primer has a nucleotide sequence selected from SEQ ID NOs 18 to 28 and another nucleotide sequence selected from SEQ ID NOs 29 to 49.
