PROBLEM TO BE SOLVED: To provide a total reflecting fluorescent microscope which enables a sliding surface to observe a molecule by targeting a dye of which oscillation plane is in an arbitrary direction. SOLUTION: This microscope successively has a laser source of a directly polarized laser beam 2, a polarization direction changing member 4 which is disposed rotatably around the optical axis of changing the polarization direction of the laser beam, a deflecting member 5 which is disposed rotatably around the optical axis changing the progression direction of the laser beam, a condenser lens 6, an objective lens 7 and a total reflecting surface 8 of the laser beam. This deflecting member exists at a rear focal plane of the condenser lens, the rear focal plane of the objective lens exists in the position of a focal plane 9 of the condenser lens, the total reflecting surface exists in the position of the focal plane of the objective lens and the laser beam transmitted to the incident surface of the laser beam on the condenser lens has a total reflecting region of a doughnut shape which totally reflects the laser beam on the total reflecting surface. The laser source having a circularly polarized laser beam and a quarter-wave plate is equally well.
