摘要 A new metalloprotease containing a thrombospondin domain (MPTS protein) is useful to treat aggrecan associated disease including rheumatoid arthritis and osteoarthritis : Independent claims are also included for the following: (1) a nucleic acid molecule (N1) encoding a metalloprotease containing a thrombospondin domain (MPTS protein) which encodes: (a) a polypeptide comprising the 959, 947, 1690 or 947 amino acid sequences fully defined in the specification, or a polypeptide having at least 35% to one of those sequences; (b) comprises the 2879, 3263, 5338 or 4086 nt sequence fully defined in the specification, or a sequence having at least 75% to one of those sequences; (c) comprises a sequence whose complement hybridizes to (a) or (b) under stringent conditions; and (d) is a fragment comprising the functional domain of (a), (b) or (c); (2) a nucleic acid complementary to N1, preferably an antisense nucleic acid molecule; (3) an expression cassette comprising the one of the above nucleic acids under control of a transcription initiation region and a transcription termination region; (4) a vector comprising the above nucleic acid and/or expression cassette; (5) a host cell transformed with the above nucleic acid, expression cassette or vector or a cell arising from that cell; (6) producing an MPTS protein; (7) a polypeptide (P1) encoded by the above nucleic acid or produced by the above method; (8) an antibody or its fragment or derivative specific for P1; (9) a hybridoma producing the above antibody; (10) identifying a modulator of MPTS, comprising contacting P1 with a substrate in the presence of a candidate and determining P1 activity; (11) identifying an agonist or antagonist of P1, comprising producing P1 from cells expressing P1, contacting P1 with a sample containing a candidate agonist/antagonist and determining inhibition or activation of binding P1 binding activity; and (12) identifying a P1 binding partner, comprising contacting P1 with a binding partner and determining P1 activity.
