EFFECTIVE METHOD FOR INTRODUCING POLYNUCLEOTIDE TO SILKWORM OVA

PROBLEM TO BE SOLVED: To provide an effective method for introducing a polynucleotide to silkworm ova and to provide an effective method for preparing a transgenic silkworm using the method. SOLUTION: The method uses a DNA introducing system capable of easily inserting a glass capillary into a hole for DNA introduction on the surface of the ova perforated by a tungsten needle through lever operation of a manipulator having integrally mounted tungsten needle and glass capillary which are separately operated by separate manipulators so far. This invention provides also a method which fixes ova on a slide glass arranged in one direction. The introduction of DNA to an objective spot in the ova becomes easy and allows increasing of the survival rate of the transgenic silkworm to ten times by the conventional method by using the above system and method.
