PROBLEM TO BE SOLVED: To provide a method for measuring protease in which a protease substrate film is used, capable of preventing a biological sample from falling even when the protease substrate is degraded by the action of the protease, and capable of surly conducting the degradation of the substrate and the observation of cell nuclei, etc., in the biological sample at the same time. SOLUTION: This method for measuring the protease includes processes (1) and (2) as follows: (1) the biological sample mounted on a flat plate is contacted with the film which contains the protease substrate and a cross-linking agent and is formed on a supporting medium in such a state as to be separable; and (2) the supporting medium is separated from the film, and then a digestion trace formed on the film by the action of the protease is detected.
