VARIANT ACTINOPLANES TEICHOMYCETICUS VAR JIANESIS AND PREPARATION OF TEICOPLANIN THEREFROM

PURPOSE: Provided is a method of purifying and preparing antibiotics teichplanin in a high yield from variant actinoplanes teichomyceticus var jianesis. More particularly, a culture medium and a culturing method are provided to maximize the production of teichplanin from the same variant strain. CONSTITUTION: The method of preparation comprises: a step of slant culturing actinoplanes teichomyceticus var jianesis(SIIA 92-363) by inoculating it to a slant medium at 30deg.C for 10 days; a step of first stage seed culturing the slant cultured strain by inoculating to the first seed culture medium at 28deg.C for 48 hours; a step of second seed culturing by inoculating to the second seed culture medium at 28deg.C for 24 hours; and a step of inoculating the seed cultured strain to nutrition fermentation medium at 28deg.C for 100 hours. The purification method comprises: a step of filtering the culture medium after making it basic; a step of extracting the filtered culture medium after it is absorbed to porous resin; a step of preparing unpurified teichplanin by concentrating the extract under pressure, crystallizing and filtering; a step of decolorizing the unpurified teichplanin by using activated charcoal after dissolving it in de-ionized water; and a step of recrystallizing the bleached product.
