摘要 PURPOSE: Provided are a recombinant vector pEST3 including thermostable esterase gene derived from Sulforobus solfataricus recombinant, its transformant and recombinant thermostable esterase produced by the transformant. Also, provided is a method for preparing optically active carboxylic acid using the recombinant thermostable esterase. CONSTITUTION: The recombinant thermostable esterase is produced by DNA recombination using the esterase gene derived from Sulforobus solfataricus. It has characteristics as followed: (a) its optimum temperature is 60-80 deg.C, (b) its optimum pH is 5.0-8.0, (c) its molecular weight is 30-34 kDa, (d) it is inhibited strongly by paraoxon and phenylmethylsulfonyl fluoride, and (d) it is stable at 60 deg.C or more for a long time with optimum activity. It has the amino acid sequence of the SEQ ID NO:1. It is used for the production of optically active carboxylic acid including (S)-ketoprofen, (S)-naproxen, (S)-ibuprofen or (S)-suprofen.
