An enzymatic compsn. is claimed comprising at least one alpha-amidating enzyme (AAE) capable of catalysing, in the presence of oxygen and a reducing agent, the conversion of a peptidyl cpd. having a glycine residue at least at the C-terminus of its peptide chain to a corresponding peptidyl amide cpd. having an amino gp. in place of the C-terminal glycine, the enzymatic compsn. being sufficiently pure in AAEs to exhibit a specific activity of at least 25 mll per mg of protein present and the enzymatic compsn. being sufficiently free of proteolytic activity capable of degrading at least one of the peptidyl cpd., the corresponding peptidyl amine and the AAEs to allow the compsn. to catalyse the conversion when the peptidyl cpd. comprises L-amino acids. Also claimed are immunoaffinity columnss for purifying the AAE and method for prodn. of AAE compsns. using DNA sequences coding for AAE.
