A pharmaceutical composition comprising an analgesic compound and a xanthine derivative in a ratio of 1 - 20: 1, preferably 10:1. ACTIVITY : Analgesic; synergist. Mature CFLP mice of both sexes, weighing 23 - 27 g were used in a mouse writhing test. The animals consumed a standard rodent laboratory diet and tap water except on the day of the experiment when they were starved for 16 hours prior to oral administration of the test substance and had access only to tap water. The mice were kept at 22 +- 2 [deg]C, cyclic lighting. A 1 % methylcellulose suspension was prepared from the test substances which was administered to the mice through a gastric tube in 0.1 ml/10 g body mass volume. The method of Witkin et al. (J. Pharmacol. Exp. Ther., 133, 1961, 400408) was used. The mice were placed in transparent containers 1 hour before measurement to acclimatize to the experimental conditions. On the day of the experiment they were injected intraperitoneally with 0.2 ml/animal of 0.6 % acetic acid solution, then after a 5 minutes waiting time the number of writhes was counted over 5 minutes. The pain reaction to the chemical noxia was determined in untreated animals (control) then the antinociceptive effect was expressed as a percentage of the control. The combination of S-(+)-ibuprofen with 5 mg/kg theobromine resulted in significant increase in the antinociceptive effect of S-(+)-ibuprofen. A 20 mg dose of S-(+)-ibuprofen had a 7 % antinociceptive effect 4 hours after administration. A dosage of 20 mg S-(+)-ibuprofen and 5 mg/kg theobromine had a 23 % antinociceptive effect after 4 hours. MECHANISM OF ACTION : None given. |
