PROBLEM TO BE SOLVED: To provide both a crystal of pyruvate dehydrogenase kinase(PDHK) and a high resolution structure of the PDHK. SOLUTION: The crystal of the PDHK protein is represented by a specific amino acid sequence and comprises the PDHK-2 amino acid sequence so as to start at the amino acid position 16 or a homologue, a fragment, a variant or a derivative thereof. The high resolution three-dimensional structure of the PDHK is obtained in the presence or absence of a physiological and a synthetic ligands by X-ray diffraction. Furthermore, the use of the structure is provided so as to identify, design or select a compound binding to the PDHK. In addition, the compound is identified, designed or selected by using the high resolution structure of the PDHK and a nucleotide sequence is used to obtain a crystallizable PDHK protein. |
