PURPOSE: Provided is a method for separating a cell from a virus completely using sucrose without reducing the activity of the virus, thereby effectively pure extracellular virus. CONSTITUTION: In a method for separating a cell from a virus comprising cytolysis of the cell infected with a virus, and centrifugation of the lysate to separate extracellular virus from the virus-infected cell, it is characterized by using a sucrose cushion having 2-5 fold higher concentration than that of sucrose in the lysate.
