PROCESS FOR PREPARATION OF FULL-LENGTH cDNA, AND ANCHOR AND PRIMER USED FOR THE SAME

PROBLEM TO BE SOLVED: To provide a process for preparation of a full-length cDNA, and an anchor and a primer used for the same. SOLUTION: This invention relates to a selectively amplifying method for the full-length cDNA containing a step comprising producing a cDNA/mRNA complex in which several dCMP groups are bonded to 3'-terminal end by acting a reverse transcriptase to the mRNA and separately adenylating a single- stranded anchor in which a biotin group or a phosphate group is bonded to the 3'-terminal end and a phosphate group is bonded to the 5'-terminal end; a step selecting the full-length cDNA/mRNA complex by selectively bonding the anchor to the 3'-terminal end of the cDNA of the cDNA/mRNA complex; and a step performing PCR using a primer specifically bonding to the anchor and the full-length cDNA/mRNA as a template. The process comprises only two simpler steps than the conventional process, that is, a reverse transcriptional process synthesizing a cDNA using a mRNA as a template and a process for bonding an anchor and a nucleic acid.
