PROBLEM TO BE SOLVED: To provide a method for simply detecting a gene mutation, a restric tion enzyme-recognizing sequence and a specific sequence by an amplification or by a quantitative analysis by using an in vitro reaction not requiring complex procedures such as a homogeneous measuring system, i.e., an electrophoresis, a so-called Bound/Free separation, and a measuring kit used for the above detecting method. SOLUTION: The method for analyzing nucleic acids comprises hybridizing a probe containing a signal-generating substance with a target nucleic acid, acting an endonuclease and an exonuclease or a protein having an activity of the exonuclease on it an releasing the signal-generating substance for detecting the gene mutation, the restriction enzyme-recognizing sequence and specific sequence in a high sensitivity.
