A method of highly sensitively and specifically amplifying a target nucleic acid in a sample by a DNA synthesis reaction with the use of a nucleotide containing a nucleotide analog; and a method of detecting the occurrence/absence of nucleotide polymorphism based on the occurrence/absence of a DNA synthesis reaction with the use of a nucleotide containing a nucleotide analog.
