MUTATED DNA POLYMERASE ISOLATED FROM AEROPYRUM PERNIX AND METHOD FOR PREPARING THE SAME

PURPOSE: A mutated DNA polymerase isolated from Aeropyrum pernix and a method for preparing the same are provided, therefore the mutated DNA polymerase has improved thermostability and processivity. CONSTITUTION: The mutated DNA polymerase with improved thermostability and processivity is isolated from Aeropyrum pernix and has the amino acid sequence of SEQ ID NO: 1, wherein a 514th amino acid in the amino acid sequence of a normal DNA polymerase isolated from Aeropyrum pernix is changed from asparagine to glutamine. A cDNA gene encoding the DNA polymerase having the amino acid sequence of SEQ ID NO: 1 has the nucleotide sequence of SEQ ID NO: 2. A method for producing the DNA polymerase comprises the steps of:: constructing an expression vector containing the cDNA having the nucleotide sequence of SEQ ID NO: 2; transforming Escherichia coli with the expression vector; and culturing the transformed Escherichia coli to express the DNA polymerase, wherein the expression vector is pGEX-Apn.
