The invention relates to a polypeptide which is able to specifically react with the antibodies of patients infected with HCV, the peptide sequence of said polypeptide comprising at least one sequence chosen from among SEQ IDL NO: 1 or SEQ IDL NO:2, chosen from the sequences having the immunoreactive properties of SEQ ID NO: 1 or SEQ ID NO: 2, and exhibiting, when compared to SEQ ID NO: 1 or SEQ ID NO: 2, at least one substitution of an amino acid by an equivalent amino acid, the equivalent amino acids being chosen from the following groups: Group 1:alanine, proline, glycine. Group 2: aspartic acid, glutamic acid. Group 3: histidine, lysine, arginine. Group 4: asparagine, glutamine, serine, threonine. Group 5: phenylalanine, tyrosine, tryptophane. Group 6: isoleucine, leucine, valine, methionine, and/or structurally modified. The invention also relates to the use of the said polypeptide for the detection and quantification of anti-HCV antibodies or of NS3 proteins, in a biological sample, or to obtain anti-bodies.
