PROBLEM TO BE SOLVED: To provide a separator and a separation method capable of separating and collecting a plasma component from a small amount of blood with high Ht. SOLUTION: The separator comprises a blood holding container 11 for holding diluted blood, a separation means 20 for separating a diluted plasma component from the diluted blood and a plasma component collecting container 31 for holding the separated diluted plasma component. The separation means 20 comprises hollow fibers 22 in a bundle form so as to come into contact with the diluted blood at the outer peripheral surface, in a separation container 21 communicating with the blood holding container 11. The hollow fibers 22 have a tube wall forming a number of through-holes having the size that allows passing of the plasma component and a diluent but prevents passing of blood cells, and its lumen communicating with the plasma component collecting container 31. In addition, the separator comprises a piston mechanism 40 capable of reducing the pressure of the plasma component collecting container 31 and the lumen of the hollow fiber 22.
