CELL-SPECIFIC REPLICATION-COMPETENT VECTOR

摘要

A vector showing expression and replication of a gene specifically in specif ic cells is constructed. A DNA having a gene expression controlling activity to be used in the construction is provided. Further, a therapeutic method where in the above vector is transferred into a specific tissue such as malignant tum or or cellsin vivo and allowed to express and replicate the gene is provided. T he transcriptional initiation regulatory domain of human calponin gene, which i s specifically expressed in cells, is obtained and ligated to the upstream of a virus replication-associated gene. Then the ligated product is integrated in to a virus DNA to thereby construct a replication vector showing cell-specific expression which would not act on normal cells in an adult. The thus constructed vector is transferred into malignant tumor cells to thereby selectively injury tumor cells or proliferative smooth muscle cells in tumor neovessels.
