<p>The invention concerns the analysis and detection of antigenic substances using immunological techniques commonly called blot. The inventions aims at providing an improved immunoblotting method which solves the problem of false-positives, white holes, and which enables to improve the reproducibility, sensitivity and reliability of the assay. This is achieved by the invention which proposes the performance of electrodouble blotting , which consists in: preparing the samples to be analysed in particular by neutralising proteases, separating the AgC protein target-antigens by isoelectric focusing, performing a first blot (western blot: WB), saturating the WB membrane, incubating said WB membrane with primary Abl antibodies, washing, dissociating the AgC/AbI complexes and transferring the released AbI on an electrodouble-blot (EDB) membrane, by electrophoresis and in the presence of acetic acid, saturating the EDB membrane, washing it, detection by chemoluminescence, acquiring and processing the light signals and interpreting them. The invention is applicable to the analysis of nucleic acids or proteins, in particular in screening recombinant EPO in urine.</p> |
