Pharmaceutical compositions containing as the active ingredient heterocyclic compounds represented by the general formula (|), isomers or solvates thereof, or pharmaceutically acceptable salts of them exhibit excellent TNF-a production inhibiting activity and are therefore useful in the prevention and treatment of various diseases caused by abnormal production of TNF-a: (|) wherein R1 is optionally substituted alkyl, aryl, heteroaryl, or the like; R2 and R3 are each hydrogen, hydroxyl, optionally substituted alkyl, or the like; R4 is hydrogen, optionally substituted alkyl, or the like; X is hydrogen, halogeno, hydroxyl, optionally substituted alkyl, aryl, heteroaryl, amino, or the like; and Y is oxygen or sulfur.
