PURPOSE: Provided are a method of determining blood-relationship by typing STR alleles on the X chromosome and a DNA typing kit using the same, thereby determining the blood-relationship between grandmother and daughter or sisters without father's DNA. CONSTITUTION: The method of determining blood-relationship by typing STR alleles on the X chromosome comprises the steps of: extracting DNA from a sample of a person to be tested; carrying out PCR of a short tandem repeat(STR) allele on the X chromosome using the extracted DNA as a template to obtain a large amount of STR DNA; measuring physicochemical properties of the STR DNA; and comparing the type of the STR allele, wherein the sample is at least one selected from blood, hair, saliva, semen, vagina secretion, isolated cell, tissue sample, dandruff, and bone; the STR is a part or total of one selected from the group consisting of DXS7133, DXS9895, DXS9898, DXS6807, GATA165B12, DXS6797, DXS6803, DXS6806, DXS8378, GATA144D04, DXS7132, DXS6789, GARA186D06, GARA31E08, ATA28C05, GATA172D05, DXS6804 and GATA164A09; each STR is amplified by using a pair of primers having the nucleotide sequence of SEQ ID NOs: 1 to 36, respectively; and the physicochemical properties of the STR DNA contains the size of fragment and the nucleotide sequence.
