METHOD FOR PRODUCING HERBICIDE-RESISTANT PROTOPORPHYRINOGEN OXIDASE GENE BY SITE-DIRECTED MUTAGENESIS

PURPOSE: A method for producing herbicide-resistant protoporphyrinogen oxidase gene by site-directed mutagenesis is provided, thereby producing high diphenyleter-resistant plants. CONSTITUTION: The herbicide-resistant protoporphyrinogen oxidase gene is produced by site-directed mutagenesis of protoporphyrinogen oxidase gene of Bacillus subtilis, in which Val 311 is substituted by Met-311. The herbicide-resistant protoporphyrinogen oxidase gene has the nucleotide sequence of SEQ ID NO: 1 and the herbicide-resistant protoporphyrinogen oxidase has the amino acid sequence of SEQ ID NO: 2. The recombinant vector pRSETB contains the herbicide-resistant protoporphyrinogen oxidase gene. A transformant is produced by transforming E. coli BL21-Gold(DE3)pLysS with the recombinant vector pRSETB.
