Novel insulin precursors comprising a connecting peptide (mini C-peptide) of preferably up to 6 amino acid residues and comprising at least one Pro are provided. The precursors can be converted into human insulin or desB30 human insulin. In one embodiment, the insulin precursor comprises the formula B(1-29)-X1 -Y -A(1-21), wherein X1 is a peptide sequence of 1-5 amino acid residues of which at least one is Pro and Y is a cleavage site.
