1275164 Derivatives of benzofuran- and benzothiophene-carboxylic acids CIBA-GEIGY AG 14 Sept 1970 [15 Sept 1969] 43806/70 Heading C2C The invention comprises compounds of formula and their pharmaceutically acceptable salts, wherein X is O or S, Y is H or Me, Z 1 is Cl, F, Me or MeO, Z 2 is H or Me, and R 1 , R 2 are each Me or Et. These compounds are prepared by (1) reacting the corresponding 2, 3-dihydro- 5- formylbemofuran- or -5-formylbenzothiophenecarboxylic acids (II) with P 1 COCH 2 COR 2 , or (2) hydrolysing the C 1-4 alkyl esters of compounds of the above formula (which may in turn be made from esters of II and R 1 COCH 2 COR 2 ). Intermediates also prepared are the corresponding benzofuran- and benzothiophenecarboxylic acids and their 2,3-dihydro derivatives; 7,8-dimethylcoumarin; 5-(2,4- or 2,6- dichlorobenzylidene)rhodanine; and 3-(2,4-dichlorophenyl)-2-mercaptoacrylic acid. Therapeutic compositions preferably in a form for oral administration, comprise compounds of the above formula, which have diuretic and saluretic activity.
