摘要 PROBLEM TO BE SOLVED: To obtain a stable image by a method of observing a sample in liquid using scanning probe microscope. SOLUTION: In the method of observing sample in liquid, a sample is observed by obtaining the image of the surface of the sample based on the interaction between the probe of a scanning probe microscope and the sample by bringing the probe closer to the sample in a liquid by using the scanning probe microscope provided with a cantilever carrying the probe at its front end, a light source, and a detector. A measure is taken to prevent the vaporization of the liquid in which is sample is dipped. Alternatively, a layer of an insulating liquid which is not mixed with the liquid is provided on the surface of the liquid and, at the time of observing the sample, only the front end section of the probe is put in the liquid and the other portion of the probe is covered with the insulating liquid. More alternatively, the light from the light source is projected upon the cantilever without passing through the interface between the liquid in which the sample is dipped and the atmosphere and the reflected light from the cantilever is caught in the liquid.
