A process for efficiently producing an optically active amino acid from a 5-substituted hydantoin which comprises, from a bacterium belonging to the genus Microbacterium capable of converting the 5-substituted hydantoin into the optically active amino acid, isolating a hydantoinase gene and an N-carbamyl-L-amino acid hydrolase gene participating in the conversion ability, and constructing a recombinant DNA having an elevated productivity of the target enzyme via gene amplification and elevation of the transcription and translation activities. The hydantoinase gene is typified by a DNA which has the base sequence represented by SEQ ID NO:1 in Sequence Listing and encodes a protein having a hydantoinase activity. The N-carbamyl-L-amino acid hydrolase gene is typified by a DNA which has the base sequence represented by SEQ ID NO:3 in Sequence Listing and encodes a protein having an N-carbamyl-L-amino acid hydrolase activity.
|
