摘要 PROBLEM TO BE SOLVED: To obtain a novel DNA expression vector useful for producing transformed yeast cells containing a first heterologous DNA sequence coding for a mammalian G protein coupled receptor and a second heterologous DNA sequence coding for a mammalian G protein α subunit (mammalian Gα). SOLUTION: This novel DNA expression vector comprises a first segment comprising at least a fragment of the extreme amino-terminal coding sequence of a yeast G protein coupled receptor. A second segment is positioned downstream from the first segment (in more correct reading frame) and comprises a DNA sequence encoding a heterologous G protein coupled receptor. The first and second heterologous DNA sequence is capable of being expressed and an endogenous G protein αsubunit (yeast Gα) is capable of being prevented from expressing in the obtained yeast cells. The cells are useful for screening compounds affecting the rate of dissociation of Gαfrom Gβγ in the cell.
