1278434 (1,4 - Dioxa - 8 - azaspiro[4,5]dec-8- ylmethyl)indole derivatives ALDRICH CHEMICAL CO Inc 30 Sept 1969 [2 Oct 1968] 48074/69 Heading C2C The invention comprises compounds (I) of formula wherein R<SP>1</SP>, R<SP>2</SP>, R<SP>3</SP> and R<SP>4</SP> are each H, C 1-8 alkyl, C 2-8 alkenyl, C 2-8 alkynyl, phenyl or benzyl, and a -process for making them by reacting in an inert organic solvent a 2-R<SP>1</SP>-1,4-dioxa-8-azaspiro[4,5]decane (II) with an appropriately substituted 5 - methylene - 6,7 - dihydroindol-4 (5H)-one. The latter are prepared by reacting the corresponding 5-dimethylaminomethyl-6,7- dihydroindol-4(5H)-one (III) methiodides with aqueous alkali. I may also be prepared by reacting II with a 6,7-dihydroindol-4(5H)-one (IV) and CH 2 O. III are prepared by reaction of IV with Me 2 NH 2 -HCl and CH 2 O. Pharmaceutical and veterinary compositions for oral or parenteral administration comprise compounds of the above formula, which have tranquillizing and antiemetic activity.
