METHOD OF SCREENING INHIBITOR FOR THE CHROMOSOMAL SEGREGATION FUNCTION OF BACTERIUM AND INHIBITOR OBTAINED BY THE SCREENING METHOD

It is intended to provide a method of conveniently and quickly searching for a novel and useful antibacterial agent which comprises bringing a test sample into contact with transgenic host cells obtained by the homologous recombination of a chromosomal segregation protein mukB gene on the chromosome of a bacterium by a plasmid carrying a fused gene having a fluorescence marker gene in the downstream of the mukB gene, a drug-tolerance gene and a temperature-sensitive mutation gene encoding a replication starting protein, and then observing whether or not a fluorescence focus is formed, thereby screening a substance inhibiting the formation of an MukFEB complex composed of MukF, MukE and mukB proteins and playing an important role in the chromosomal segregation. The substance obtained by this screening method, which inhibits the formation of the chromosomal segregation protein complex, is highly useful as an antibacterial agent based on a novel function mechanism.
