PURPOSE: Provided are a kit for detecting the nucleic acid of HCV with specificity and sensitivity and a detection method of HCV by using the kit. CONSTITUTION: The detection kit comprises: a RNA extract solution containing DEPC treated distilled water, Taq buffer, and DMSO; a primary PCR master mixed solution including DEPC treated distilled water, buffer, dNTP, a primer set having nucleotide sequences of SEQ ID NO:1 and 2, DTT, Taq/RNasin and reverse transcriptase; and a secondary PCR master mixed solution containing distilled water, buffer, dNTP, a primer set having nucleotide sequences of SEQ ID NO:3 and 4, Taq/UDG and cresol red. The detection method comprises the steps of: treating a sample with a RNA extract reagent and a primary PCR master mixed solution then performing PCR; treating the PCR products with a secondary PCR master mixed solution then performing PCR; and analyzing the presence of HCV on the gel.
