PROBLEM TO BE SOLVED: To obtain a thermophilic prolylendopeptidase useful for structural analysis of proteins. SOLUTION: This isolated DNA contains a nucleotide sequence encoding a polypeptide having thermophilic prolylendopeptidase activity. This recombinant vector contains the DNA. This expression vector contains the DNA. This transformant is transformed by the expression vector. This method for producing the polypeptide by using the transformant is provided. A formed prolylendopeptidase builds up an essential protein structure by heat treatment at a high temperature for a fixed time and can actualize high activity. The thermophilic prolylendopeptidase is obtained from Pyrococcus horikoshii.
