METHOD AND KIT FOR SEPARATING PLASMID DNA USING SILICATE POWDER

PURPOSE: Provided are a method and a kit for separating plasmid DNAs using silicic acid matrix prepared by treating silicate powders with strong alkali and acid. The plasmid DNA is useful for the cloning in E. coli. CONSTITUTION: The method for separating plasmid DNA comprises the steps of: obtaining cells from E. coli culture solution by centrifugation and suspending the obtained cells; performing cytolysis with the suspended cells; separating plasmid DNA from the cell lysate; precipitating the separated plasmid DNA and allowing the precipitate to bind silicic acid matrix; obtaining the plasmid DNA- binding silicic acid matrix by centrifugation and washing it; drying the obtained matrix; separating the plasmid DNA and silicic acid matrix and eluting the plasmid DNA; and separating the plasmid DNA eluate.
