摘要 A diamond-nucleic acid conjugate having a nucleic acid moiety regularly bonded at a high density, showing a strong chemical bond to a solid base, being chemically stable, being stably usable in organic solvents etc., and never inducing the deterioration in the nucleic acid. Diamond is covalently bonded to thymidine to form a diamond-thymidine conjugate. Then the thymidine structure of this diamond-thymidine conjugate is reacted with a nucleic acid and thus bonded thereto. To bond diamond to thymidine, a diamond powder is exposed to a strong acid to thereby bond carboxyl group to the surface of the diamond and then thymidine is bonded via ester-bond to this carboxyl group. Next, a nucleic acid is bonded to the thymidine under the ester-bond. As the nucleic acid, use is made of a nucleic acid having adenosine bonded to the 3'-terminus. In case of amplifying by the PCR method, adenosine is usually bonded to the 3'-terminus. Thus, adenosine, which is complementary to thymidine, may be used.
