PURPOSE: A differentiation regulating method of embryonic stem cells using a pine leaf extract(PE) is provided, thereby the differentiation of embryonic stem cells can be inhibited, so that the embryonic stem cells can be effectively cultured. CONSTITUTION: The differentiation regulating method of human or animal embryonic stem cells comprises adding 1 to 10 microgram/liter. preferably 3 microgram/liter of the pine leaf extract(PE) as leukemia inhibitory factors into a growth medium suitable for culturing human or animal embryonic stem cells and culturing the embryonic stem cells at 37 deg. C in an atmosphere of 5% carbon dioxide, wherein the pine leaf extract(PE) is produced by washing the pine leaves with water, extracting them with a mixed solvent of water and ethanol in the ratio of 1:1 at 90 deg. C for 4 hours; filtering the extract and concentrating the filtrate under reduced pressure.
