The invention relates to a method for the direct identification of target nucleotide sequences, target amino acid sequences or target antigens by the amplification of DNA using a DNA polymerase and a DNA double-strand destabilising enzyme. According to the invention, the target molecule is present in situ or in solution, or is directly immobilised, or immobilised by means of a capture probe on a solid phase. A DNA polymerase that is capable of strand displacement, dNTPs and a DNA double-strand destabilising enzyme are then added and the resultant amplification product is directly detected, the target being identified by means of said detection. To identify non-amplifiable target molecules, a coupling probe carrying a DNA matrix is first added and a DNA polymerase that is capable of strand displacement, dNTPs and a DNA double-strand destabilising enzyme are then added and the resultant amplification product is directly detected, the target being identified by means of said detection. |
