PURPOSE:To obtain in an industrially advantageous way the subject compound with high optical purity by culture of Brevibacterium microorganisms capable of producing D-alanine and resistant to beta-chloro-D-alanine followed by collecting the product from the cultured product. CONSTITUTION:Brevibacterium microorganisms capable of producing D-alanine and resistant to beta-chloro-D-alanine [e.g. Brevibacterium lactofermentum HR 5-43(FERM P-13338)] is inoculated into a medium and put to shaking culture at 30 deg.C for 16hr, and the grown microbial cells are collected and thoroughly washed with a physiological saline solution. A suspension containing the microbial cells is then inoculated into a medium containing beta-chloro-D-alanine to perform a culture at 30 deg.C. Then, the resulting culture solution is filtered and the microbial cells are produced and accumulated, and the filtrate is then centrifugalized to eliminate the microbial cells. The pH value of the resulting filtrate is adjusted to 2 and the filtrate is injected into a strongly acidic cation exchange resin followed by eluting the adsorbed substance with dilute ammonia water, and the elute is concentrated, followed by addition of ethanol to the concentrate and the objective crystal D-alanine produced is collected. |
