<p>PROBLEM TO BE SOLVED: To obtain a collagen-coated cell culture vessel or culture member capable of easily reproducing in vitro the polarity of cells in vivo. SOLUTION: This collagen-coated cell culture vessel or culture member is obtained by the following practice: a collagen solution of a specified concentration is dividedly cast and applied at 10-1,000μl/cm2 each with a pipette onto any culture vessel so that the surface thereof become uniform, and the resulting vessel is left at rest on a surface with an angle of inclination of <=45 deg. at 15-42 deg.C for 20-120 min and dried for 15 min to 72 h; alternatively, a collagen solution of a specified concentration is applied at 10-1,000μl/cm2 onto an elastic culture substrate such as a silicone film immediately followed by making a polymerization in an incubator at 15-42 deg.C, and the resulting elastic culture substrate is left to stand for 15 min to 72 h under a UV lamp in a clean bench, and after the collagen is dried, the resultant culture substrate is moistened again with a phosphate buffer solution and then stretched at 10-40% and fixed.</p> |
