PROBLEM TO BE SOLVED: To provide a means capable of simply removing calcium, magnesium, etc., from a polyester-based cloth to an extent not affecting analysis even if the cloth comprises ultra fine fibers of 1 denier or less in monophyletic fineness in the case of a manufacturing method of an improved integral-type multilayer analysis element, for example, used for analysis of calcium, magnesium, etc., in aqueous liquid specimens, specifically, in living body fluids such as blood. SOLUTION: This method is used for preparing an analysis element having a reagent layer containing an indicator showing an optically detectable change by bonding to objective ions and a spread layer comprising a polyester cloth, in this order, on a light-transmitting, water-impermeable base. The manufacturing method of this integral-type multilayer analysis element is characterized by cleaning the polyester cloth with a water solution containing an organic solvent before it is layered.
