The invention provides a therapeutic antisense technology based on snap back stealth oligonucleotide to activate a highly cytotoxic substance within target cells of interest but not within normal host cells. Activation of the toxin depends upon hybridization of the antisense strand with the target nucleic acid. A partially homologous or pseudosense strand modified with a toxin such as Mitomycin. A becomes single stranded and random coiled. The toxin adducts which have been hydrogen bonded to an antisense strand and inert in a double stranded molecule are active in a single stranded, random coiled molecule. The toxin such as Mitomycin A reacts with cell components that cause cell toxicity and cell death. |
