An in vivo method of producing a Product nucleic acid molecule comprising: (a) introducing into a host cell (i) an Insert Donor nucleic acid molecule, comprising a desired nucleic acid segment flanked by a first recombination site and a second recombination site, wherein the first and second recombination sites do not recombine with each other; and (ii) a Vector Donor nucleic acid molecule, comprising a third recombination site and a fourth recombination site, wherein the third and fourth recombination sites do not recombine with each other; (b) causing the Insert Donor and Vector Donor nucleic acid molecules to recombine within the host cell, thereby producing a Product nucleic acid molecule; and (c) isolating a host cell comprising the Product nucleic acid molecule but not the Insert Donor nor the Vector Donor nucleic acid molecules. The method enables efficient and specific recombination of DNA segments using recombination proteins. The DNAs, vectors and methods are useful for a variety of DNA exchanges, such as subcloning of DNA in vivo.
