An enzyme electrode, characterized in that it comprises an electrode system comprising an insulating substrate and, formed thereon, a working electrode and a counter electrode and optionally a reference electrode, and a reaction layer comprising a diaphorase (DI), 12a-hydroxysteroid dehydrogenase (12a-HSD) and nicotinamide adenine dinucleotide synthetase (NADS), wherein at least a part of the reaction layer is positioned on the working electrode and DI,12a-HSD and NADS contained in the reaction layer are immobilized on the surface of the working electrode so that a compound generated in the reaction layer can reach the surface of the working electrode; a method for determining the concentration of adenosine triphosphate (ATP) using the enzyme electrode; and a system for carrying out such determination.
