METHOD FOR THE PREPARATION OF SELECTIVELY RANDOMISED NUCLEIC ACID MOLECULES

We describe methods for producing selectively randomised oligonucleotides using a phosphoramidite dinucleotide and mononucleotide synthesis strategy. These methods comprise: (i) coupling a dinucleotide phosphoramidite to the 3' position of a nucleotide; (ii) coupling a mononucleotide phosphoramidite to the oligonucleotide of (i); and (iii) repeating steps (i) and (ii) until the desired length oligonucleotide is produced. This methodology provides randomised oligonucleotides without the problems of NNN randomisation, without having to resort to complicated resin-splitting procedures or the use of low coupling efficiency trinucleotide phosphoramidites.
