PURPOSE: Provided is a manufacturing method of activated carboxypeptidase Y(CPY), effectively inhibiting the generation coagulation while producing CPY in E.coli. CONSTITUTION: Activated CPY is effectively mass-produced by over-expressing CPY propeptide and CPY in transformed E.coli BL21(DE3)/pEHCPYPR(KCTC18026P) and E.coli BL21(DE3)/pECPYM(KCTC18025P) then using produced CPY propeptide as an adjuvant in the refolding reaction of CPY.
