PROBLEM TO BE SOLVED: To obtain both a liquid culture medium for culturing acid-fast bacteria capable of effectively preventing contamination to cause a problem when the acid-fast bacteria are cultured in a liquid culture medium and shortening the test period while maintaining the acid-fast bacteria at the detection efficiency equal to or higher than the conventional detection efficiency and a solid-liquid two-phase culture medium for culturing the acid-fast bacteria comprising a combination of a solid culture medium with the liquid culture medium in order to adapt to a sample pretreating agent for culturing the acid-fast bacteria consisting essentially of cetyl pyridinium chloride(CPC). SOLUTION: This modified M7H9-B culture medium for selecting and separating the acid-fast bacteria is characterized as comprising hemin, L-asparagine hydrate, casein peptone, a yeast extract and oleic acid in a basal culture medium comprising M7H9-B culture medium. The two-phase culture medium comprising the combination of a solid Vite culture medium with the liquid modified M7H9-B culture medium in order to adapt to the sample pretreating agent comprising the CPC is obtained.
