Method involves culturing Clostridium histolyticum in casein-soybean-pepsin medium containing the following components, g/l: casein and soybean oil cake hydrolyzate, 958,8-979,2; sodium dihydrogen phosphate hydrate, 1.4-1.6; potassium hydrogen phosphate dihydrate, 1.4-1.6; pyridoxine, 0.0019-0.0021; riboflavin, 0.0019-0.0021; thiamine bromide, 0.0019-0.0021; folic acid, 0.0019-0.0021; calcium pantothenate, 0.0019-0.0021; nicotinic acid, 0.0019-0.0021; lipoic acid, 0.0019-0.0021; biotin, 0.0009-0.0011 and distilled water, the balance, to 1 l. Purification and concentration of enzyme are carried out using ultrafiltration membranes with retention threshold value of substances 50 kDa by molecular mass. Invention can be used in medicine in therapy of burn and infected wounds, in perfume industry and in scientific investigations, for example, for determination of collagens structure. Invention provides to develop the simple, easily scalable and ecologically pure process for isolation and purification of collagenase from Clostridium histolyticum.
