Polypeptide (I) with beta -secretase activity that can cleave specifically the natural precursor (APP) of the beta -amyloid peptide (bA). Independent claims are also included for the following: (a) a compound (Ia), that is not (exclusively) a peptide and able to cleave APP at the beta -secretase site, produced by reproducing the active motifs in (I) with non-peptide structures; (b) production of (I) in a human cell line (specifically THP1, ATCC TIB 202) that is representative of the central or peripheral nervous system or the immune system and is able to metabolize normal APP; (c) detecting compounds able to inhibit interaction between (I) and its substrate, using the cells defined in (b); (d) antibody (Ab), or its fragments, directed against (I) and able to inhibit interaction between (I) and APP, inhibit activity of (I) and/or interferes with metabolism of bA. |
