The present invention relates to a method of producing a protein of interest comprising allowing a polynucleotide fusion construct to be expressed in a plastid thereby to produce a fusion protein comprising the protein of interest, wherein the fusion construct comprises a polynucleotide coding sequence of the protein of interest operably linked to a polynucleotide coding sequence of a fusion protein partner and wherein the fusion protein thus expressed has a greater half-life than the individually expressed protein of interest, is resistant to internal cleavage in vivo and comprises a cleavage site.
