PROBLEM TO BE SOLVED: To provide a power transmission device of an engine for motorcycle capable of preventing the enlargement of a dimension in the longitudinal direction. SOLUTION: In this power transmission of the engine for motorcycle provided with a V belt type transmission mechanism 8 connecting a drive side pulley 42 mounted in an outer end part of a crankshaft 7 with a driven side pulley 43 mounted in an outer end part of a main shaft 9 arranged at the back of the crankshaft 7 by a V belt 44 and a chain type transmission mechanism 12 connecting a drive shaft 15 arranged at the back of the main shaft 9 with a rear wheel shaft 90 by a chain, the V belt type transmission mechanism 8 is arranged on one side of a crank case 2, the chain type transmission mechanism 12 is arranged on the other side of the engine and at the back of the crank case 2, and the main shaft 9 is arranged on an upper side more than a straight line joining the drive shaft 15 to the rear wheel shaft 90.
