PROBLEM TO BE SOLVED: To provide a method of fixing a single stranded DNA fragment on a substrate surface by conjugation it using a disulfonic compound and a detection method of a DNA chip and a nucleic acid fragment as a sample. SOLUTION: In the method for fixing a single stranded DNA fragment having a reactive group on a substrate surface at a terminal part thereof, a double helical DNA fragment is bonded onto the substrate surface through a bond group: -R3R2C-R1HC-SO2-L-SO2-L-SO2-CHR1-CR2R3, a bond group: -R3R2C-R1HC- SO2-L-SO2-(CR1R2)n- or a bond group: -(R2R1C)n-SO2-L-SO2-(CR1R2)n-[wherein R1, R2 and R3 represent atom or group independent of each other selected from among a group comprising 1-6HC alkyl group and 6-20 Calyl group, n integer of 1-6 and L bivalent combination group].
