A biosensor which has a developing layer comprising a reagent fixing section (5) for fixing an antibody against a substance to be measured in a solution to be tested and a reagent holding section (4) for holding the above antibody being in a dry and labeled state and eluted by the development of the solution to be tested and which determines the substance to be measured in the solution to be tested qualitatively or quantitatively by measuring the amount of the labeled reagent bound to the reagent fixing section (5), characterized in that the side surface of the developing layer which is parallel with the direction of permeation of the solution to be tested has been partly or wholly hardened and closed through melting; and a method for preparing the biosensor. The biosensor allows appropriate arrangement and adjustment of the permeation of the solution to be tested in the developing layer and also allows provision of a precise biosensor at a low cost.
